Computing area around centerline points

I segmented the coronary artery and implement centerline algorithm. Now get the end points of centerline. But now i want to get the distance between the endpoint and nearest neighbouring . can anyone tell me by using which function i can get the distance in MATLAB???

Second question is if i want to compute the area at one pixel location.....you can say with first endpoint then how it is possible in matlab??
