Optical flow between two regions in two images

Let's say I have the coordinates of several points that define a region in image 1 (I1). I also have the coordinates of several points that define a region in image 2 (I2). These regions are not in the same locations in the two images, and their areas are not necessarily the same. Is there any way to calculate optical flow between these given regions in the two images?
