Coding a height in a Pipe from a series of images

Hi I'm looking for codelines which may be of use to me.

I need to analyse the height of beads flowing within a small tubular pipe.

Ideally I believe I need to greyscale a series of images (to allow them to be more visible and measurable). Then take two points on each image and measure the changing height of both points (which remain constant through each image on the x axis; but change through the y) through 1000+ images.

If anyone has any good codelines which may be of help I'd be really appreciative as im tring to jigsaw pieces together.
