Method to determine the difference between 2 images

I need suggestion on how to differenstiate the diffetence between this 2 images. The help will be much more appreciate =)

There are lots of things that are different between those images, like size, area, count, and shape of dark blobs. See my Image Segmentation Tutorial in my File Exchange
