Retina Blood vessels Segmentation

I have task to do blood vessels segmentation from fundus images, the images used in this task was given our collaborators in this task. i need to segment into three regions which are 1. background 2. tumor 3. blood vessels . after preprocessing stage i obtain the images attached in this question, but how to the results from gold standard ( manual segmented image)
