After creating a gui from GUIDE, there will be two files, one is .fig and the other is .m.
I want to know and learn how to get the gui programmatically, so I need to know the code in .fig file. Is this possible?

You don't need to delve into the .fig file because the .m file has everything you need. If you really want to get into the tedious business of writing your own GUI creation code from scratch, you can look at 41-complete-gui-examples.
But I really recommend you use GUIDE or App Designer because for anything but the simplest user interface, you're going to have millions of controls to create, with billions of properties to specify (OK, maybe an exaggeration), and it will be very time consuming and tedious.

In all recent MATLAB, there is no code in .fig files.
What will be there is graphics objects, some of which will have callbacks set to property values such as
@(hObject,eventdata)YourGuiNameHere('calculate_Callback',hObject,eventdata,guidata(hObject))
