how to further subdivide a subplot

I have setup four subplot areas using subplot(2,2,1), subplot(2,2,2), subplot(2,2,3) and subplot(2,2,4).

Is it possible to further divide subplot(2,2,4) to subplot(2,1,1) and subplot(2,1,2) ?

Also is it possible to create a pop up dialog box that mentions "updating graphics - this may take a few minutes" whilst calculations are progressing prior to graphics updating and automatically disappears when processing has completed and graphics have displayed.
