Automatic Registration reverse vectors

I am interested in procuring an algorithm than can look at a serial section stack and calculate the reverse alignment vectors between each slice, and enter these vectors into a matrix.

THe serial stack will be intensity only images.

These vectors can be rotational and/ or translational.

I don't think I catch how this would be different than standard image registration ?

There are some algorithms that would execute the transformation, that is, perform the registration, but not output the rotation or translation vector, eg -34 degrees or -56 x or 900 y!
