You pressed the Run button to execute the code. When you press the Run button, MATLAB executes the code with no parameters, and makes no attempt to look anywhere to see if there just happens to be a variable named image_in.

You need to go to the command line and invoke the function, passing in the file name. Or do the equivalent in another block of code m
